Mechanochemical preparation of chrysomycin A self-micelle solid dispersion with improved solubility and enhanced oral bioavailability

Abstract Background Chrysomycin A (CA) has been reported as numerous excellent biological activities, such antineoplastic and antibacterial. Though, poor solubility of CA limited its application in medical field. Due to good amphiphilicity potential anticancer effect disodium glycyrrhizin (Na 2 GA) an excipient, amorphous solid dispersion GA/CA-BM) consisting Na GA was prepared the present study by mechanochemical technology (roll mill ML-007, zirconium balls, 30 rpm, 2.5 h) improve oral bioavailability CA. Then, GA/CA-BM self-assembled micelles water. The interaction state were investigated X-ray diffraction studies, polarized light microscopy, scanning electron microscope. Meanwhile, properties sample solution analyzed dynamic scattering transmission electron. Furthermore, antitumor ability vivo tested, providing a theoretical basis for future on cancer therapy. Results encapsulated nano-micelles average diameter nano-micelle 131.6 nm, zeta − 11.7 mV. Three physicochemical detections showed that transformed from crystal into form after treated with ball milling increased 50 times. Na2GA/CA-BM significant increase about two time free Compared CA, in-vivo studies also exhibited had inhibition tumor growth. Conclusions nanoparticles (131.6 mV) simple low-cost can efficacy vivo, suggesting formulation efficient treatment. Graphic abstract
